Abstract
To continue, and extend into a new area, our research on the eukaryotic cytosolic chaperonin, CCT, we propose to use a combined molecular- and cell-biological approach to: (1), clone and provide molecular biological characterisation of cDNAs encoding Physarum CCT subunits and (2), use the unique cell biology of Physarum, including the exquisite natural synchrony of its cell cycle, to study the biochemistry of CCT function in relation to cytoskeletal remodelling, to mitotic spindle formation and other cell cycle related events, to centrosome-directed microtubule assembly and to flagellum construction. In this way, cellular functions of the individual subunits of this essential molecular chaperone will be defined.
