Abstract

The androgen receptor (AR) is a ligand-activated transcription factor that regulates genes important for male sexual development and differentiation. The identification and study of protein-protein interactions between the receptor and the general transcription machinery will be central to an understanding of the mechanism(s) of receptor-dependent gene regulation. Recently, work by the applicant identified the basal transcription factor TFIIF as one target for the human AR transactivation domain. However, this work did not delineate the step in the transcription reaction regulated by the AR. The present proposal will further characterise the interaction between the AR and TFIIF and exploit cell-free transcription systems to investigate the mechanism(s) of AR-dependent gene activation. In addition, novel target proteins binding to the AR will be screened for. The objective of these studies will be to gain a better understanding of the mechanism(s) of AR action.
