Abstract

Two 1998 Nature papers identified a caspase-activated deoxyribonuclease (CAD) together with an inhibitor (ICAD) which regulated the activity of CAD in apoptosis. Within the predicted amino acid sequence of ICAD but apparently unnoticed by the original authors, two hallmark motifs of an aspartic proteinase are present, suggesting that ICAD may be a precursor form of an aspartic proteinase and so may well contain intrinsic proteolytic activity. This proposal requests pump-priming funds in order to provide unequivocal proof of this concept and thus indicate whether such an activity may contribute to the complexity of proteolytic events that mediate apoptosis.
