Abstract

The dynamic internalisation and recycling of G protein-coupled receptors (GPCRs) is enhanced by agonist ligands and is intimately involved in the desensitisation and resensitisation of cellular response. Using green and blue fluorescent protein-linked constructs of the long and short isoforms of the thyrotrophin releasing hormone receptor, and mutations of these receptors which are know or are likely to regulate their degree of internalisation and constitutive activity, we will provide a detailed molecular description of the trafficking pathways of GFP-tagged GPCRs, real time information on the mechanics of receptor movement in intact cells and an analysis of the contributions of signalling pathways to these processes.
