Abstract
The Adamalysins are transmembrane proteins with roles in adhesion and the activation of cell-associated factors. In addition to extracellular metalloproteinase and disintegrin domains, several ADAMs, including TNF-alpha converting enzyme (TACE, ADAM-17) possess cytoplasmic motifs that may link them to signalling pathways regulated by tyrosine kinases. With ADAM-17 as a model, we will use molecular techniques to study ADAM cytoplasmic domain-interacting proteins in macrophages and the roles of ADAMs in signalling. We will: (1) identify interacting proteins using GST-ADAM fusion proteins; (2) study the effects of macrophage activation on association and phosphorylation status; (3) analyse ADAM involvement in inside-out signalling; (4) devise dominant-negative strategies to ablate ADAM- mediated signalling.
