Abstract

Protein 4.1 (p4.1) is a component of the plasma membrane-associated cytoskeleton in most animal cells. Despite being the subject of investigation in erythrocytes for many years, its role in complex tissue is still poorly understood. This project will investigate the role and regulation of p4.1 in neurons, using the P19 cell line as a model. P19 mouse embryonal carcinoma cells can be differentiated to neurons, accompanied by changes in p4.1 isotype expression. To determine the nature and roles of individual p4.1 isotypes, their mRNAs will be characterised and their expression inhibited by anti-sense oligonucleotides. High molecular mass isotypes of p4.1 are rapidly induced at synaptogenesis, and are components of post-synaptic densities. p4.1 interactions with PSD proteins in vitro will be analysed. A 65 kDa isoform of p4.1 which interacts only weakly with the cytoskeleton will be investigated.
