Abstract
The aims of this project are: (1) to determine the structure of the p19/Cdk6 complex using X-ray crystallography; (2) to attempt cystallisation of cyclin D/Cdk6 both on its own and with inhibitors; (3) to attempt to express and purify Cdc37, a putative Cdk4/6 chaperone protein, to study its interactions with Cdk6 and, if appropriate, to set up crystallisation trials of that complex, and (4) to use the structural information in an attempt to understand the mechanisms of inhibition of the cyclin D/Cdk6 complex by the Ink4 proteins.
