Abstract

Dedicated technical support is requested to underpin an active and successful programme of work aimed at elucidating the structure of photosystem two. Electron microscopy is being used to analyse ordered 2D crystals and single particles of PSII in various states of complexity (in terms of subunit content and functional activity). Moreover, it is planned to initiate a series of 3D crystallisation trials for X-ray analyses of various PSII complexes and proteins. The new focus is to improve the reproducibility of the biochemical preparations from spinach and exploit new starting materials, such as Chlamydomonas and Synechococcus, in order to drive the work to higher resolutions. Growth and culturing facilities are therefore also requested.
