Abstract

The synthesis of lipids containing differential levels of cis unsaturated versus saturated fatty acids lies at the heart of the phenomena of chilling resistance in plants. Diffraction quality crystals of Squash Glycerol-3-phosphate-1-acyltransferase (G-3-P-AT), the enzyme which catalyses the incorporation of the acyl group from acyl-Acyl Carrier Protein or acyl CoA's into glycerol-3-phosphate, has been obtained and this proposal aims to determine its three dimensional structure. Once completed the structure will be analysed against the sequence of the highly homologous Arabidopsis enzyme to provide new insights into the molecular basis of its specificity for cis-unsaturated fatty acids versus saturated fatty acids and to enhance our understanding of factors influencing substrate specificity and thereby frost sensitivity.
