Abstract

Genetic engineering has been employed to modify the primary processes of photosystem II. Residues which have been targetted are D1-130 and D1-198, which we have already shown to modulate the free energy and quantum yield of the primary radical pair in the isolated PS II reaction centre, and D1-His118, a potential ligand to a putative linker chlorophyll between the reaction centre and antenna chlorophylls of PS II. Time resolved spectroscopy will be used to determine the effect of these mutations upon the energy and electron transfer within PS II core complexes and complimented by determination of in vivo growth rates. These studies are aimed at elucidating the balance between efficient function and regulation in this photosystem, with the long term objective of identifying potential targets for genetic manipulation of PS II for improved photosynthetic efficiency/stress tolerance.
