Abstract

We and others have shown that physiological levels of nitric oxide (NO) reversibly inhibit mitochondrial respiration at cytochrome oxidase, and that cells stimulated by cytokines to express iNOS regulate cellular respiration by this means. This project aims to determine whether NO is a physiological or pathophysiological regulator of cellular respiration and its affinity for oxygen, by testing whether and in what conditions endothelial agonists or cytokines causing NO release from ecNOS or iNOS in cultured endothelial cells, cultured vascular smooth muscle cells or isolated aorta can regulate the cellular respiration of these and surrounding cells via NO inhibition of cytochrome oxidase or peroxynitrite inhibition of the respiratory chain. Whether the known effects of NO and cytokines on cell growth, division, and death are mediated via the inhibition of respiration will also be tested.
