Abstract

Flavocytochrome P-450 BM3 is an extremely important enzyme. This enzyme from Bacillus megaterium is the only soluble P-450 to use a mammalian-like diflavin P-450-reductase. In addition, the reductase domain is covalently linked to the P-450 haem domain providing a one-component system for study. We aim to provide an integrated thermodynamic and kinetic profile of this important enzyme by completing the following objectives: Determining the reduction potentials for all three cofactors in the enzyme (FAD, FMN and Haem); performing a thorough kinetic analysis of each electron transfer step in the enzyme; and mutating key residues to determine their effects on the redox characteristics of the enzyme.
