Abstract
Fab fragments of monoclonal antibodies to MHC Class II molecules will be covalently bound to small fluorescent particles (phycobiliproteins and latex beads). These probes will be bound to receptors on monocyte/macrophages and imaged using a cooled CCD camera. Receptor mobility will be analysed by sequential imaging and receptor clustering by single particle image analysis techniques. The effects on these properties of stimulation by selected cytokines and antigenic peptides will be investigated. The functional implications of redistribution of MHC Class II molecules in antigen presenting cells will be explored.
