Abstract
It has been suggested that Ca transport by the Ca-ATPase involves four Ca binding sites, two on the cytoplasmic side of the membrane, and two on the luminal side. Chemical labelling will be used to identify residues making up the luminal sites, employing reconstituted lipid vesicles and a novel fluorescence assay. Affinity methods will be used to purify ATPase labelled at luminal sites. Rapid kinetic methods will be used to study effects of binding at these sites on phosphorylation. Site-directed mutagenesis will be performed on key residues.
