Abstract
Recent work highlights our ignorance of the lignin biosynthetic pathway and suggests that alternative routes may exist for the synthesis of some lignin precursors. We want to investigate this further by genetic manipulation, blocking the pathway at multiple positions to elucidate the existence of alternative biosynthetic routes. Down-regulation of multiple genes can be difficult to achieve. We suggest a strategy where linked partial sense sequences for multiple genes are expressed from a single promoter. Our aim is to determine the universal utility of this strategy and to use it to produce a series of lignin mutants for in-depth study.
