Abstract
We will use an electrophysiological approach to characterise the interaction of Ryania alkaloids with purified sheep cardiac sarcoplasmic reticulum Ca2+-release channels. The influence of the alkaloids on single-channel gating and conduction will be monitored under voltage-clamp conditions following the incorporation of channel proteins into planar phospholipid bilayers. The data produced from the study will provide information on the structural features of the alkaloids which govern interaction with the receptor domain of the channel protein, the location of the binding domain within the channel protein and the mechanisms underlying the functional consequences of the interaction.
