Abstract

The aim is to elucidate the 3D structure and molecular mechanisms of the important membrane proteins for transport of glucuronide-(H+, Na+) and L-fucose-H+ found in bacteria. Specific questions are: which parts of the proteins recognise sugar and/or cation; which parts contribute to trans-membrane sugar-cation movement; is the 12-helix model of the protein structure true? The principal techniques are 2D and 3D crystallisation trials on each protein, and electron or X-ray diffraction. The mechanism will be studied by protein engineering, allied to biochemical, genetical, immunochemical, and biophysical methods.
