Abstract

1. To locate those amino acid residues governing specificity/selectivity for fatty acid and hydrocarbon substrates and those determining kinetic properties by site-directed mutagenesis of P450 BM3. 2. To prepare and characterise the functional sub-domains which comprise the reductase of P450 BM3 by cloning and overexpressing the sub-genes in E. coli. 3. To determine the microscopic rate constants for P450 BM3 and its constituent domains and sub-domains by stopped flow kinetics. 4. To attempt to crystallise P450 BM3 and its reductase domain.
