Abstract

Fluorescence ratio photometry and imaging techniques will be used to study the kinetics, generation and maintenance of oscillations, and putative waves, in cytosolic free Ca2+ ([Ca2+]i) in stomatal guard cells in response to external Ca2+ ([Ca2+]e). The ability of two plant hormones, abscisic acid and auxin, and temperature to modulate oscillations in [Ca2+]i in response to [Ca2+]e will be investigated. The contribution of [Ca2+]e and intracellular Ca2+ stores to the oscillatory behaviour will be studied, and the signal transduction elements involved in the generation and maintenance of oscillations will be examined using caged probes and tobacco transformed with antisense constructs of known signal transduction genes.
