Abstract
A dramatic reorganisation of the actin cytoskeleton takes place on pollen activation and germination. We believe we have a clone encoding a protein, named ZmABP1, which is involved in this process. The deduced amino acid sequence of ZmABP1 predicts an actin and phosphoinositide binding protein which shows considerable sequence similarity to several small actin depolymerising proteins. We will examine the binding of recombinant ZmABP1 to F-actin, G-actin and phosphoinositides. We will determine the effects of re-ZmABP1 on the polymerisation/depolymerisation of actin at different pH values and in the presence of phosphoinositides using a co-sedimentation assay. We will follow the spatial distribution of ZmABP1 from pollen to germinated pollen using immunolocalisation microscopy. The effects of increasing the level, by microinjection, of ZmABP1 on cytoplasmic organisation and the actin cytoskeleton in pollen tubes will be investigated. In addition, a minor component of this project will be to characterise ZmABP1 like clones isolated from a maize pollen cDNA library.
