Abstract

P-glycoprotein is a unique protein, associated with both active transport and volume-regulated chloride channel activities. These two functions are separable and appear to represent alternate functional configurations of the protein. The aims of this proposal are threefold:to investigate the mechanism of interconversion between the transporter and channel configurations, particularly the role of protein kinase C; to elucidate the physiological role(s) of the P-glycoprotein associated chloride channel in epithelial physiology (particularly of the gut); to study the functional relationship between the P-glycoprotein channel and the closely related epithelial chloride channel protein CFTR (the product of the cystic fibrosis gene).
