Abstract
The aim of the present proposal is to investigate how the transition between the cellular and scrapie forms of the prion protein propagates between neural cells. This will be investigated in systems where identified populations of cells are confronted in dissociated culture. The work should give information about the timing and rate of intercellular infection, and its consequences for target cells at the cellular and molecular level. The analysis will encompass primary glial cells from the mouse PNS (Schwann cells) and CNS (astrocytes), and possibly nerve cells. The work should have general implications for agents which inhibit propagation.
