Abstract

The aim of this proposal is to purify and characterise the hexosephosphate translocator (HPT) of wheat endosperm amyloplasts. The purified protein which is capable of transporting glucose 6-phosphate and glucose 1-phosphate will be incorporated into liposomes and its transport characteristics studied, particularly with respect to the stoichiometry of hexosephosphate/inorganic phosphate counter exchange. Attempts will be made to obtain full length clones of HPT from a wheat endosperm cDNA library. Site directed mutagenesis will be undertaken to determine the relationship between structure and substrate specificity of the translocator which will be expressed in yeast.
