Abstract
The total synthesis of the metal-containing cofactors Moco of the oxomolybdoenzymes and Tuco of the tunzymes is proposed. The synthesis of molybdopterin (MPT) is underway at Manchester but the synthesis of metal-oxo-complexes with one and two MPT per metal remains a major challenge. Strategies for this coordination chemistry will be developed and products characterised structurally, electrochemically and spectroscopically (e.g. XAS, EPR, ENDOR, ESEEM, NMR). The synthetically-derived cofactors will be incorporated into Mo and W apoproteins, especially for isotopic labelling of MPT for spectroscopic studies of intermediates in the catalytic cycle. Isolation of MPT trapped by a suitable metal centre will be attempted for full characterisation.
