Abstract

We have cloned, over-expressed and purified the gene regulatory protein AreA, and shown that the protein binds specifically to a DNA sequence from the A. nidulans genome. We now wish to (i) determine the range of DNA operator sites to which AreA binds, and probe the protein-DNA interactions at these sites (ii) initiate studies of the 3-D structure of AreA and appropriate DNA-protein complexes (iii) investigate the mechanism of transcriptional repression by measurements of the affinity of AreA for specific sites and the modulation of DNA binding by co-factors.
