Abstract

Plant RAR1 that contains two zinc-binding CHORD domains mediates disease resistance against various pathogens. These CHORD domains associate with components of the protein folding- and degradation-machinery including SGT1 and HSP90. This grant aims to determine the structure of each CHORD domain and intact RAR1. Furthermore, we will elucidate interaction surfaces and other molecular details of the SGT1-CHORD-II interaction and the HSP90-CHORD-1 interaction. We will also assess whether an HSP90-RAR1-SGT1 ternary complex forms and whether the structure of RAR1 and its affinity for partner proteins changes during resistance activation.
