Abstract

In collaboration with Professor Claudia Daubenberger, Unit Leader of Clinical Immunology at the Swiss Tropical and Public Health Institute, Basel, we will conduct a pilot study on 100 peripheral blood mononuclear cell (PBMC) samples before and after vaccination, from the Group’s ongoing vaccination studies in Tanzania. These samples are already available and have been quality-controlled. We will first test the efficacy of extracting RNA and DNA from these samples. We will then generate RNA-seq and VDJ-seq sequencing libraries which will be run on the Illumina Hiseq at the Babraham Sequencing Facility. We will perform flow cytometry analysis and use Flowsom modelling of flow cytometry data to deconvolute changes in PBMC populations. We would aim to build on this pilot in subsequent applications by applying the refined approach to additional cohorts from different ODA-recipient countries, and treated with different vaccines.
