Abstract

Experimental manipulation of intrinsic stem cell factors can be used to regulate self-renewal and differentiation in mouse ES (mES) cells. With the development of readily propagateable hES cells we can now consider performing analogous studies in human cell lines. The aim of this project is to identify intrinsic regulatory factors (RFs) that play a functional role in controlling hES cell fate. We have recently demonstrated that transient transfection of short interfering RNAs (siRNA) can efficiently knock-down gene expression in hES cells. We will therefore apply this approach to screen for functionally relevant genes in hES cells. Expression of a select pool of candidate stem cell specific regulators identified in published data bases will be knocked down using RNAi and the consequences on stem cell self-renewal and growth will be monitored. The identification of functionally relevant hES cell regulators will increase both our understanding of stem cell biology and improve the efficiency with which we can produce medically relevant cell types.
