Abstract

Polymorphism in the major histocompatibility complex (MHC) modulates immune responses through variable binding of antigenic peptides. We have developed a model system for MHC class II structure-function analysis using cattle characterised for distinct immune responses to a peptide antigen (FMDV15). The influence of MHC polymorphism on the T cell responses seen in these cattle will be analysed using transfectants and transgenic mice expressing cloned class II DR and DQ genes . In related projects we will be mapping the cattle MHC, and investigate variation in MHC polymorphism and immune response to vaccination in a large crossbred resource herd (RoBogen). These studies will allow the contribution of MHC and other loci to be evaluated in a relevant cattle population.
