Abstract

Plant (tobacco) assimilatory nitrate reductase will be overexpressed in yeast and large quantities will be prepared for structural spectroscopic and mechanistic characterisation. This will include analysis of reductive and oxidative half reactions, steady-state and pre- steady-state kinetics and binding of substrates and inhibitors. Methodology will include UV/visible, EPR and ENDOR spectroscopies and X-ray structure determination if possible. Directed mutants will be prepared to investigate hypotheses about the function of various amino- acid residues.
