Abstract

The main aims of this project are to add SAR, actin terminator and lox sequence to the expression cassettes; Evaluate affect of adding introns to the luciferase ORF; Transformation of wheat using constructs pAL250 and pAL253; Characterization of the TO parent; Determination of luciferase transgene copy number and quantification of its expression in single seed derived populations; Determine if genetic background significantly alter expression; Mapping transgene loci using the chinese spring monosomics; In situ hybridization to localize position of transgene on chromosome; Recovery of transformed plants from transformed callus.
