Abstract
The proposed work will develop a series of binary and super- binary vectors based On the Mullineaux binary vectors to study the transfer of T-DNA to regenerable callus of wheat and barley. T-DNA transfer will be quantitated by time-lapse video-imaging. The affects of SAR elements, introns and reversed termination signals on enhancement of expression. reduction of position effects and gene silencing respectively will be studies in single seed derived populations using quantitative RT-PCR.
