Abstract
We will develop and evaluate vectors for the large-scale functional characterisation of plant genes by activation tagging in Arabidopsis. A panel of related vectors are proposed for a) rapid generation of derivative loss-of-function alleles from activation-tagged alleles, b) conditionally enhanced expression of tagged alleles and c) suppression of closely related members of gene families. We will test the rationale underlying each vector in reconstruction experiments in planta and evaluate rapid mapping of T-DNA insertion sites in tagged lines by sequencing of TAIL-PCR products. The utility of these vectors in functional genomics will be evaluated by characterisation of several thousand activation-tagged lines for each vector.
