Abstract
We aim to determine the ultrastructural organization of active RNA polymerase I transcription complexes by a combination of different microscopies (confocal, TEM, tilted view reconstruction, TEM of replicas). We will determine their structures both in their native conformation in well-preserved (rapidly frozen) cells and at differing degrees of disruption by hypertonic and physical spreading, in order to arrive at a detailed picture of the native, active conformation.
