Abstract
This is a supplementary award to grant 208/IGF12436 in which resources will be provided for systematic analysis of gene function in S. coelicolor. Transposon libraries containing random insertions will be phenotypically characterised and the disrupted genes identified. PCR based procedures will be developed for large- scale targeted mutagenesis by gene replacement with linear cassettes. Phage based vectors for insertional mutagenesis will be developed and exploited to insert regulated promoters to control essential genes, and to transduce insertions to form multiple mutants. Detailed functional analysis will focus on putative regulatory genes. All resources and methods will be made widely available, together with access to a co- ordinated database of all mutants, linked to existing S. coelicolor databases. (Joint with grants 58/IGF12431 and 208/IGF12432.)
