Abstract
We will provide resources for systematic analysis of gene function in S. coelicolor. Transposon libraries containing random insertions will be phenotypically characterised and the disrupted genes identified. Linear cassettes generated by PCR will be used for targeted gene replacement. Phage-based vectors for insertional mutagenesis will be developed and exploited to insert regulated promoters to control essential genes, and to transduce insertions to form multiple mutants. Detailed functional analysis will focus on putative regulatory genes. All resources and methods will be made widely available, together with access to a co-ordinated database of all mutants, linked to existing S. coelicolor databases.
