Abstract
The clean gene technology to be developed in this project uses existing Agrobacterium-mediated procedures and employs a newly developed highly flexible binary Ti vector system which has been funded by the BBSRC. This approach relies on the co- transformation into plants of two T-DNAs, only one of which contains the selectable marker gene. In a proportion of ransformants, the integration of the T-DNAs into separate loci will occur. Subsequently, a self-crossing of the transformant will lead to segregation of the marker T-DNA from aT-DNA carrying a test gene. The project will determine the best parameters for achieving the maximum frequency of segregation of the T-DNAs, provide a flexible plasmid system to encourage use, techniques to eliminate primary transformants with linked T-DNAs prior to transfer to containment glasshouse and a demonstration of its use in two UK crops.
