Abstract
The aims of this project are to generate data that complement the needs for assembling the E. tenella genome and, from those data, derive a more comprehensive understanding of the organisation of individual chromosomes that, in conjunction with analyses of the progeny of a genetic cross, inform on the outcomes of recombination during the sexual phase of reproduction. The objectives of the project are to: 1. Extend the mapping of representative genes on the molecular karyotype to determine their chromosomal locations. Genes chosen initially will be those identified as comprising specific metabolic pathways. 2. Feed these data in to the assembly of the genome at the Sanger Institute. 3. Following on from previous studies by Rahmat Sheriff, focus on the organisation of chromosomes 3 and 4 ¿ a comprehensively assembly for these chromosomes will be expected to aid in the identification of regions that contribute to size polymorphisms. 4. Through collaboration, examine synteny between chromosomes of E. tenella and the sequences of other apicomplexans within the public domain. 5. Analyse the inheritance of markers in independent genetic crosses in E. tenella between the pathogenic Wey strain and the WisF96 hyper-abbreviated line and selection of recombinant progeny.
