Abstract
The objectives of this project are: 1) To investigate the upregulation of proapototic markers caspase-3 and Fas throughout the incubation period of disease in the ME7/CV mouse model and compare the results with those obtained in the 87V model. 2) To compare the upregulation of poly(ADP-ribose)polymerase (PARP) one of the proteins cleaved in the caspase pathway, and its relationship with caspase-3 and Fas in both the ME7/CV and 87V/VM model. 3) To develop a FACS analysis technique to measure and compare pro-apoptotic markers and cell types involved in the apoptosis observed in the brains of mice in both the ME7/CV and 87V/VM mouse models. 4) To relate any apoptotic changes observed in both mouse models to the cytoskeletal dysfunction observed in both mouse models.
