Abstract

There is a need to generate new tools to study porcine lymphocyte populations. Traditionally, monoclonal antibodies would be raised against porcine lymphocytes and screened for those that show specificity for cells of immunological interest. The target antigen would then be characterised using biochemical methods. We will take a different approach in this project and exploit the growing information available in public databases to isolate porcine genes of immunological interest, and use these to generate antibodies from expressed protein. We have shown recently that porcine lymphocyte populations contain high levels of cells which resemble natural killer (NK) cells. Antibodies that recognise porcine NK receptors are not available at present, but sequences of the genes encoding the subunits of the porcine NKG2D/DAP receptor are available in the public database. This information will be used to isolate c-DNA¿s encoding these receptors for use in protein and antibody production. Molecular techniques will also be used to generate porcine MHC class 1 tetramers (see P0788). Again database sequences will be used to isolate c-DNAs encoding porcine MHC class 1 genes and these will be used to generate recombinant protein for tetramer construction.
