Abstract

Our work is to model on how virus persists in foetal tissue. Our model uses infection of early passage tissue culture cells that show a wide range of innate immune responses with BVDV. This programme of experiments is directed towards determining the roles of individual virus polypeptides in establishing persistent infection, and dissecting the responses of the cell to infection with virus having such an unusual replication strategy. Work will be directed to understanding how the virus avoids the response of cells to dsRNA. We postulate that the glycoproteins of the virus may be able to interfere, in some way, with the initiation of the signalling cascade that leads to interferon stimulated gene expression. We have developed insect cell-based systems for the expression of secreted forms of each of the glycoproteins and these will be utilized to test this hypothesis. In addition, influence of other virus polypeptides in this and other cellular processes will be investigated.
