Abstract
Dendritic cells are the only antigen presenting cell considered capable of initiating primary T cell responses in naïve animals. In cattle ¿ex-vivo¿ dendritic cells can be isolated from the afferent lymphatics draining the skin. A number of sub-populations of these dendritic cells are evident, as defined by staining with monoclonal antibodies. Ex-vivo cells will be compared with dendritic cells derived from precursors in blood by culture with cytokines that provide a model for less mature cells. Differences in the synthesis of cytokines and co-stimulatory surface molecules by dendritic cell sub-populations that will influence the bias of the T cell response are evident. Synthesis of cytokine transcripts and protein by the dendritic cell sub-populations and expression of established and novel co-stimulatory surface molecules by dendritic cell sub-populations will be determined. The response of subsets of bovine CD4, CD8 and gamma delta TCR+ T cells to antigens presented by the dendritic cells and the effect on the bias and type of the T cell response will be assessed. This will enable the manner in which the dendritic cells and T cells interact and respond to bacterial, viral and model antigens and how this has the potential to influence resistance to disease to be examined.
