Abstract
ASFV encodes a number of proteins that interfere with the host response to infection by modulating signalling pathways in infected macrophages. One of these proteins, A238L, has dual functions; it inhibits activation of the host transcription factor NFkB and inhibits calcineurin phosphatase activity. The aims of the project are to understand how these dual functions of A238L protein are regulated. Specifically the role of nuclear/cytoplasmic shuttling and post-translational modification of the A238L protein in the regulation of A238L function will be investigated. This will involve defining how the A238L protein is modified post-translationally and determining where this occurs and what effect this has on A238L function. The distribution of different molecular weight forms of the A238L protein in the nucleus and cytoplasm at various times after infection of cells with ASFV will be investigated and the interaction of A238L with NFkB and calcineurin in nucleus and cytoplasm studied. Putative nuclear localisation and nuclear export signals in the A238L protein will be investigated by in vitro mutagenesis and effects of mutating these signals on the distribution and function of the protein studied.
