Abstract

To generate molecular data that will establish the population structure and biology of the bovine pathogen S.uberis, by exploiting technological developments in rapid nucleotide sequence determination and analysis, and provide a rational framework for subsequent studies. S.uberis isolates, from representative samples of populations, collected in the UK over the last 6 years will be characterised by direct nucleotide sequence determination of loci encoding housekeeping proteins. This approach will also be applied to isolates of the closely related species S.parauberis. In addition, the project has the following secondary objectives: A. To clarify the importance of the putative virulence determinant PauA, the plasminogen activator. The presence/absence of the gene (pauA) that encodes this protein, and subsequent diversity at the molecular level, will be explored in isolates highlighted in the first part of the study. Results from this will be correlated with the data from the MLST study, providing informed strain selection for subsequent studies; and B. To identify/characterise novel virulence determinants of S.uberis. Representational difference analysis will be used to identify potential genomic differences between well-characterized strains of S.uberis, identified in the initial parts of the project, that differ in their ability to cause clinical mastitis. This approach will lead to the identification of novel virulence factors, which may be involved in the ability of a S.uberis strain to cause disease.
