Abstract

Pestiviruses have three structural glycoproteins (Erns, E1 and E2). Both Erns and E2 bind to the surface of cells. Erns binds to cells through interactions with cell surface glycosylaminoglycans and E2 probably interacts with a specific receptor for the virus. Other functions of the glycoproteins, such as cell fusion activity, have yet to be assigned to a specific glycoprotein. The aims of the project are to define parameters of virus entry into cells: to describe any conformational changes in the glycoproteins that may take place during virus entry into cells; to examine the consequence of any change in conformation on the properties of the glycoproteins; and to investigate the route of virus entry into cells. It is hoped that a new way to purify virus from tissue culture will be developed which we hope will aid these analyses of the precise role of the virus glycoproteins in virus entry into cells.
