Abstract
The objectives of the project are: 1. Identification of NFkB and calcineurin binding sequences encoded within African swine fever virus gene A238L in order to develop corresponding peptoid analogues with clinical applications. 2. Finish/complete sequencing of complete Ectromelia virus (EV) genome. 3. Identification of novel viral modifiers of apoptosis, TNF and cytokine responses from the sequence of EV. 4. Identification of novel viral modifiers of apoptosis and TNF and cytokine responses through screening cloned viral ORFs and expression libraries of EV and mouse gamma-herpes virus 68 (MHV) 5. Identification of possible human homologues of novel viral modifiers of apoptosis, TNF and cytokine responses. 6. Identification of cellular targets of ASFV, EV and MHV modifiers of apoptosis, providing novel targets and strategies for development of drugs controlling cell proliferation. 7. Manipulation of apoptosis and TNF in vitro with virus by transgenic expression of virus host evasion genes, in order to provide new strategies for controlling immune responses in health and disease and to produce cell lines with improved growth characteristics for biotechnology. 8. Construction of virus deletion mutants as novel vectors for DNA delivery, protein expressions and vaccines. 9. Provide structural information on viral evasion molecules leading to design of new peptoid drugs for manipulation of immune responses
