Abstract
Probing conformational changes in the structure of prion proteins using nucleic acid ligands. RNA ligands for the prion protein have been developed in collaboration with Oxford University and the student is investigating the interactions of these molecules with wild-type and truncated/mutated forms of purified recombinant prion protein by gel shift analysis, analytical ultracentrifugation and various spectroscopic methods. The objective is to obtain high resolution structural data on the prion:RNA complex, to understand at the atomic level the basis of the specificity of this interaction and to use this information to guide a structure-based molecular design approach for better ligands. Such ligands have potential for diagnostic and therapeutic applications in the TSE field.
