Abstract

We have recently characterised three monoclonal antibodies that bind to cells showing a distribution and morphology typical of FDC in sheep and cattle. The proposed project will attempt to further characterise the molecules recognised by the monoclonal antibodies using a variety of molecular biological and protein chemistry techniques e.g. constructing and screening expression libraries, protein purification and sequencing. Immunising mice with purified cells will also be used to extend the panel of monoclonal antibodies available. Functional characterisation of the different populations of putative FDCs from different anatomical sites will be performed in vitro in B cell proliferation and antibody production assays. In addition, the capacity of the cells to bind and retain antigen will be examined in longitudinal studies using immunisation with whole virus and recombinant viral proteins. Analysis will be performed using immunohistochemistry, flow cytometry and infectivity assays.
