Abstract

The recently developed MHC class I tetramer technology will be adapted and optimised for use with cattle MHC class I genes. A number of characterised genes and peptides will be assessed during the first stage of the project. The resulting reagents will be used to quantitatively and qualitatively analyse T cell responses in cattle to respiratory syncytial virus, an important pathogen of young animals. This technique is unique in that it allows direct isolation of MHC and peptide-specific T cells from peripheral blood and tissues. It will be possible to study the kinetics, longevity and functional characteristics of antigen specific T cells following infection with RSV. In addition it will allow evaluation of vaccines under development.
