Abstract

The objectives of this project will be to analyse the chicken NK region in a systematic manner with the aim of identifying the genes present in this region in chickens so that their individual roles functions can be analysed. This will involve constructing a set of bacterial artificial chromosome (BAC) clones across this region, identifying genes within the BACs and investigating polymorphism and patterns of expression for selected genes. Initial BAC and YAC clones isolated from this region have confirmed the genetic mapping data relating the chicken and mammalian regions and allowed mapping of the chicken clones against the existing human BAC map of the region. As the chicken contig is extended sample genes identified in these BACs will allow their alignment with the corresponding human BAC map and make it possible to predict which chicken BAC(s) span the NK Ly49 and NKRP1 clusters. This will be confirmed by limited sequence skimming of the BACs, and exon trapping, before commercial sequencing of the confirmed BACs using funds available from an EU project. Analysis of the sequence of the region will allow detailed comparison of the NK gene structure numbers and organisation with that of humans and mouse, and will also allow a limited set of genes of particular interest to be selected for detailed investigation of expression levels and polymorphism in birds of different genotypes and resistance status.
